Melanotan II and Tanning

Melanotan has demonstrated significant effects on melanin production (melanogenesis), also known as “sunless tanning” or skin tanning without sun exposure. It does so by binding primarily to MC-1R and MC-4R, and has lesser effects in MC-3R, three of the five melanocortin receptors in the body. MC-1R is found in melanocytes in the skin and is primarily involved in the darkening of the skin, and helps the skin produce melanin. In contrast, MC-3R is involved in appetite and impulse control, and MC-4R is involved in sexual arousal, libido, and male erectile functionality.

It is critical to note that long-term side effects of sunless tanning, including the use of melanotan II peptides, have not been established, and that treatment with prolific agents like MT-2 may potentially stimulate the proliferation of existing skin cancer.

Melanotan II and Receptor Expression

MT-2 was found to decrease fat storage and hunger behavior in animal models. Research suggests that MT-2 has similar effects to the hormone leptin and could be effective for reducing hunger by stimulating both leptin-dependent and leptin-independent pathways. Both MT-2 and leptin are thought to cause an increase in thyrotropin-releasing hormone (TRH) gene expression and the MC-4R pathway, stimulating a region of the brain associated with satiety and food intake.
