Selective androgen receptor modulators (SARMs) have been suggested as potential therapies for addressing breast cancer, muscle wasting, and urinary incontinence in females. Although the androgen receptor (AR) is present in the uterus, the effects of SARMs on uterine function remain uncertain.

Study Design

A mouse model was employed to compare the influence of SARMs (Andarine S4, Enobosarm), Danazol, and dihydrotestosterone (DHT) on tissue structure, cell proliferation, and gene expression. 

Ovariectomized mice received daily treatment with the compounds or vehicle control (VC) for seven days.

High-throughput image analysis was used to quantify uterine morphometric characteristics, while immunohistochemistry and RT-qPCR were employed to evaluate protein and gene expression, respectively.

Results

Treatment with GTx-024, Danazol, or DHT resulted in significant increases in body weight, uterine weight, and the surface area of the endometrial stromal and epithelial compartments compared to the VC. GTx-007 treatment did not impact these parameters. 

GTx-024, Danazol, and DHT all substantially increased the percentage of Ki67-positive cells in the stroma, while only GTx-024 affected epithelial cell proliferation.

GTx-007 significantly enhanced the expression of Wnt4 and Wnt7a in the uterus, while GTx-024 and Danazol decreased their expression.

Conclusion

In conclusion, the effects of GTx-024 and Danazol on uterine cells resembled those of DHT, whereas GTx-007 had minimal effects on the parameters examined.

This study has identified specific endpoints that highlight discrepancies in the impact of SARMs on uterine tissue and establishes a framework for preclinical investigations comparing the effects of compounds targeting the AR on endometrial function.

In the future, the cycle length of therapy on these mice should be increased to improve the validity and repeatability of the findings.

Andarine S4 and Lung Cancer Prospects

Lung cancer is a leading cause of cancer-related deaths worldwide, with adenocarcinoma being the most prevalent subtype. Diagnosis involves imaging studies and tissue biopsy, and treatment options depend on factors such as cancer stage, type, and patient health. 

Advances in targeted therapies and immunotherapies have improved lung cancer treatment.

Current therapies include surgery, radiation therapy, and systemic therapy, which encompasses chemotherapy, targeted therapy, and immunotherapy. However, limitations such as treatment resistance and toxicity persis.

Andarine, a member of the SARMs class, exhibited anti-growth activity, suppressed migration, and induced apoptosis in lung cancer cells. Gene expression analysis revealed modulation of genes associated with cell cycle progression, apoptosis, and senescence.

Andarine treatment increased senescence-associated β-galactosidase activity, suggesting accelerated senescence. Cellular senescence is a response to stress that can suppress tumor growth. Andarine upregulated genes associated with senescence and cell cycle arrest, providing evidence for increased senescence rate.

Overall, the findings highlight the potential of SARMs, such as andarine, as novel therapeutic agents for lung cancer by targeting AR signaling and modulating gene expression associated with cancer cell proliferation, apoptosis, and senescence.

Andarine Research Studies

Andarine is less prevalent on the research end compared to Ostarine and Ligandrol. Currently, there are no documented clinical human trials using Andarine S4.
